高压氧预处理对皮瓣移植术后大鼠缺血再灌注损伤炎症反应的影响 被引量:3

Effects of hyperbaric oxygen preconditioning on the levels of neutrophil elastase and high mobility gooap box 1 in rats following skin flap transplantation

摘要 目的探讨高压氧(HBO)预处理对皮瓣移植术后大鼠缺血再灌注损伤炎症反应的影响。方法56只SD大鼠按数字表法随机均分为7组:假手术组(SH组)、缺血再灌注1d组、3d组、5d组(IR1d组、3d组、5d组)、HBO预处理+缺血再灌注1d组、3d组、5d组(HBO+IR1d组、3d组、5d组)。建立腹部带蒂移植皮瓣动物模型,进行HBO预处理,应用ELISA法检测各组大鼠血清中性粒细胞弹性蛋白酶(NE)及高迁移率族蛋白B1(HMGBl)水平。结果IR1d组、3d组和5d组NE水平明显高于SH组(P〈0.01),HBO4-IR1d组NE水平明显低于IR1d组(P〈0.01),HBO+IR3d组和5d组NE水平低于相应的IR组(P〈0.05)。IR1d组、3d组和5d组HMGBl水平明显高于SH组(P〈0.01);HBO+IR1d组、3d组HMGBl水平低于相应IR组(P〈0.05);HBO+IR5d组HMGBl水平明显低于相应IR组(P〈0.01)。结论HBO预处理可使皮瓣移植术后大鼠NE和HMGBl水平降低,减轻移植术后缺血再灌注损伤造成的炎症反应,提高皮瓣成活率。 Objective To investigate the effects of hyperbaric oxygen (HBO) preconditioning on inflammatory reaction induced by ischemia-reperfusion (IR) in rats following skin flap transplantation. Methods Fifty-six Sprague-Dawley rats were randomly divided into 7 groups : the sham group, the ischemia-reperfusion (IR) day 1 group, the IR day 3 group, the IR day 5 group, the HBO preconditioning + ischemia -reperfusion day 1 group, the HBO + ischemia-reperfusion day 3 group, and the HBO + ischemia-reperfusion day 5 group. The abdominal pedicle skin flap animal model was established and the animals received HBO preconditioning. The levels of neutrophil elastase ( NE ) and high mobility group box 1 ( HMGB1 ) of 7 animal groups were measured with ELLSA. Results The levels of NE in the IR day 1, day 3 and day 5 groups were obviously higher than those of the sham group. The NE level of the HBO preconditioning + IR day 1 group was obviously lower than that of the IR group (P 〈0.01 ), and the NE levels of the HBO preconditioning + IR day 3 and clay 5 groups were lower than those of the IR group( P 〈 0.05 ). The levels of HMGB1 in the IR day 1, day 3 and day 5 groups were significantly higher than that of the sham group (P 〈0.01 ). The HMGB1 levels of the HBO preconditioning + IR day 1 and day 3 groups were lower than those of the IR group (P 〈0.05). The HMGB1 level of the HBO preconditioning + IR day 5 group was obviously lower than that of the IR group ( P 〈 0.01 ). Conclusions Experiments showed that HBO preconditioning could decrease the levels of NE and HMGB1 inrats following skin flap transplantation, alleviate inflammatory reaction induced by ischemia-reperfusion following skin flap transplantation, and increase survival rate of the grafted skin flap.
