摘要
目的 :该方法可否作为一种检测药物胚胎发育毒性效应的手段 ;方法 :取孕9 5d(头褶中期 )的大鼠胚胎 ,分别在大鼠血清培养基和人血清培养基 (90 %的人血清 ,1 0 %的大鼠血清 ,并补充葡萄糖到 3mg ml血清 )中培养 48h ,并用Klug等人的评分系统对胚胎生长参数和形态进行评分 ;结果 :9 5d的大鼠胚胎在人血清培养基中培养48h ,全部胚胎均发育正常 ,与在大鼠血清培养基中培养 48h的胚胎生长发育情况相近 ;除胚胎DNA含量以外 ,其他各项评价胚胎生长发育的指标与在纯大鼠血清中培养的胚胎比较均无显著性差异 (P <0 0 5 ) ;结论 :在我们的实验条件下 ,以人血清为培养基培养大鼠胚胎的方法可用于胚胎早期生长发育的研究和评价。
Objective:To study the growth and development of 9.5 day rat embryos in whole embryo cultures,and if also may be used as a methed for testing the effects of exposure to drugs on embryos development;Methods:Growth of head fold stage(9.5 day)rat embryos cultured for 48 hours in human serum by supplemented with glucose (3mg/ml serum),and 10% rat serum.The comparison of the embryo development in rat serum,and evaluated 48 hours later for alterations in growth and specific moropholagical parameters,using a modifiod Klug scoring system;Results:The embryos growth and development in human serum is as good as that in rat serum,there are not growth retardation or abnormal development,except DNA contents of embryos cultured with rat serum,there were no statistically significant differences between data the two groups;Conclusion:The head-fold-stage(9.5 day)rat embryos cultured in human serum may be used for assessment of growth and development of embryos,it also may be used as a methed for testing the effects of exposure to drugs on embryos development.
