摘要
目的探讨原花青素(GPC)在体外对人白血病细胞(HL-60细胞)增殖抑制及细胞凋亡的诱导作用。方法体外培养HL-60细胞,分别用100、200、300、400、500μg/mL GPC作用。MTT法检测不同浓度GPC作用24、48、72h后HL-60细胞的增殖抑制率,流式细胞术检测24h后HL-60细胞的凋亡情况。结果随着GPC药物浓度的增加,其对HL-60细胞的增殖抑制率逐渐增高,呈时间-剂量依赖效应关系;GPC在低浓度(200μg/mL)时可引起少量的细胞发生凋亡,随着GPC浓度的增加,细胞凋亡率也在不断增加,呈明显的量效关系。结论 GPC可在体外抑制HL-60细胞的增殖,并诱导HL-60细胞凋亡。
Objective To probe the effects of procyanidins inducing inhibition of proliferation and apoptosis in human leukemia cells(HL-60 cells) in vitro. Methods Human leukemia HL-60 cells were respectively cultured with different concelatrations of procyanidins(100,200,300,400 and 500 μg/mL). We detected the proliferation of HL-60 cells after 24,48 and 72 h by MTT methods. HL-60 cells were cultured with different concentrations of procyanidins for 24 h,and their apoptosis were determined through flow cytometry. Results The growth inhibitory rate of HL-60 cells increased along with the concentration of procyanidins and it had the concentration and time dependent; Treated by 200μg/mL procyanidins for 24 h could induce apoptosis of HL-60 cells, the apoptosis rate of HL-60 cells increased along with the concentration of procyanidins in an apparent concentration-effect dependant manner. Conclusion Procyanidins can inhibit proliferation and induce apoptosis of HL-60 cells in vitro.
