摘要
目的应用同工酶技术研究不同种及居群栀子的亲缘关系和遗传多样性。方法采用聚丙烯酰胺凝胶不连续垂直板电泳系统,测定了12份不同居群栀子的过氧化氢酶(POD)、酯酶(EST)同工酶酶谱。结果 12份供试材料在2个同工酶系统中均有栀子物种的共同特征谱带,同时又蕴涵着丰富的多态性。结论栀子居群间的遗传变异较大,而居群内部的分化程度很低。不同居群的栀子与种的特性和地理分布具有一定相关性。该结果为栀子的品种鉴定和合理利用提供了实验依据。
Objective To determine the genetic diversity and genetic relationship in different species and populations of Gardenia jasminoides Ellis by isoenzyme technique. Methods Two isozymes (POD and EST) of 12 different Garde nia jasminoides Ellis populations were studied by polyacrylamid gel electrophoresis and cluster analysis with NTSYS 2.10 software. Results The basic features of the two isoenzyme bands were observed and polymorphism was also detected in the 12 specimens. The enzyme spectra of POD and EST were significantly different. Conclusion Genetic variations exist among populations with very low differentiation inside populations. The features in different populations show relation between species and geographical distribution, Which offers experimental evidence for identification and available application in germplasm resources of Gardenia jasminoides Ellis.
