摘要
目的检测大鼠缺血肺组织中microRNA(miRNA)表达变化,寻找与肺缺血相关的miRNA。方法将Wistar大鼠随机分为假手术组和缺血组,每组6只。缺血组参照改良Eppinger方法建立大鼠肺缺血模型(缺血1h)。建模1h后取材提取RNA,应用miRNA芯片技术检测两组大鼠肺组织中miRNA的表达。结果同假手术组相比,缺血肺组织中有4个miRNA表达上调≥2倍,21个miRNA表达下调≥2倍。结论在肺缺血过程中,miRNA的表达发生明显变化,可能对缺血肺组织中细胞凋亡起着重要的调节作用。
Objective To detect the expression of microRNA in rats with acute pulmonary oligemia. Methods Wistar rats were randomly divided into sham operation group and ischemia group, with six rats in each group. A model of pulmonary oli- gemia wa.~ created by using modified Eppinger method (one-hour ischernia). Samples for RNA extraction were collected one hour after the establishment of the model, the expression of RNA was detected in both groups by employing microRNA microarray chip. Results Compared with the sham-operation group, four pieces of miRNA up regulated more than two times in ischemic rats, and 22 pieces were down regulated more than twice. Conclusion In the course of pulmonary oligernia, the expression of microRNA changed obviously, which probably plays a important regulation on apoptosis in lung tissue with ischemia.
