摘要
目的:建立生物样品中人参炔醇(panaxynol)的高效液相色谱(HPLC)测定方法。方法:通过对精密度、特异性、回收率、稳定性等考察建立了测定方法,得到标准曲线。结果:生物样品中的panaxynol浓度(C)与色谱峰面积(A)均呈现良好的线性关系,回归方程的相关系数r均大于0.99;panaxynol测定方法的精密度高,重现性好,血浆和各组织样品测定的日内精密度和日间精密度RSD都小于10%;回收率较高,满足生物样品回收率在80~120%的范围,相对标准偏差均小于6%;稳定性好,-20℃冷贮放置不影响测定结果。结论:本法的稳定性、精密度、回收率、专属性和定量线性范围均达到了体内药物分析的要求。
To establish methodology of biological samples determination of panaxynol by High Performance Liquid Chromatography (HPLC). Methods : The determination methods were estabishded through expedition of accuracy, specificity, recovery, stability, the standard curve was received. Results: The concentration of panaxynol (C) in biological samples and the chromatographic peak area (A) showed a good linear,the correlation coefficient of regression equation (r) were greater than 0.99 ; The determination of panaxynol had high precision, good reproducibility,The day precision and inter-day precision RSD of determination in plasma and various tissue samples were less than 10% ; recovery rate was high to meet the biological samples in the range of 80 to 120% ,relative standard deviations were less than 6% ; sta- bility were good, - 20℃ cold storage did not affect the determination of the results. Conclusion: The stability, precision, recovery, specific and quantitative linear range reached in vivo drug analysis requirements.
