摘要
目的 探讨多药耐药基因产物P糖蛋白 (P glycoprotein,P gp)和P5 3蛋白在胃肠道腺癌中协同表达的意义。方法 应用免疫组织化学方法检测术前未进行化疗的 2 5 9例胃癌和 14 0例结肠癌组织中P gp和P5 3蛋白的表达。结果 P gp和P5 3在胃癌中的表达分别为 2 6 2 5 % (68/2 5 9)和3 7 0 7% (96/2 5 9) ;在结肠癌中的表达分别为 2 3 5 7% (3 3 /14 0 )和 4 0 % (5 6/14 0 )。P gp表达与胃癌和结肠癌的组织学分级、浸润深度和淋巴结转移状况无关 (P>0 0 5 ) ;而P5 3表达与淋巴结转移有关 (P <0 0 0 1) ,与组织学分级和浸润深度无关(P >0 0 5 )。P gp在P5 3阳性病例中的表达明显高于P5 3阴性的病例。结论 P gp和P5 3常协同表达于胃癌和结肠癌组织中 ,并可作为胃肠道腺癌临床耐药和预后判断的一个可靠指标。
Objective To investigate the correlation of the co expression of P glycoprotein (P gp) a multidrug resistance gene product and P53 protein in gastroenteric adenocarcinomas. Methods Two hundred and fifty nine patients of gastric carcinoma and 140 patients of colonic carcinoma,without undergoing preoperative chemotherapy, were studied for the expression of P gp and P53 protein using immunohistochemistry. Results P gp and P53 were detected in gastric carcinoma in 26 25%(68/259) and 37 07%(96/259), and in colonic carcinoma in 33/140(23 57%) and 56/140(40%), respectively. No correlation was observed between P gp expression and histological grading, invasive depth and lymph node metastases of the tumors ( P >0 05) and nor the correlation between the P53 expression and the histological grade,invasive depth of the tumors ( P >0 05),except for the lymph node metastases ( P <0 001) in gastroenteric adenocarcinomas. The P gp expression was significantly higher in P53 positive than in P53 negative tumors ( P <0 001). Conclusions The co expression of P53 and P gp is a good predictive factor for chemoresistance and for prognosis of patients with gastroenteric adenocarcinomas.
