中华圆田螺纤维素酶提取及纯化工艺的优化 被引量:2

Optimization of Extraction and Purification Technology of Cellulase from Cipangopaludina cahayensis

摘要 以中华圆田螺(Cipangopaludina cahayensis)为原料提取纤维素酶,采用均匀设计优化提取工艺,并采用硫酸铵沉淀法初步纯化酶液。结果表明,中华圆田螺纤维素酶提取最佳工艺为缓冲液pH 4.8、浸提时间12 min、离心时间24 min;采用饱和度45%的硫酸铵沉淀法纯化酶液,所得酶液的总酶活和比活力最高,分别为651.01 U和34.099 U/g。 Cellulase was extracted from Cipangopaludina cahayensis. The extraction conditions was optimized by uniform de- sign; and the cellulose extracted was purified by ammonium sulfate precipitation. The results showed that the optimum condi- tions for cellulase extraction were buffer pH 4.8; extraction time, 12 min; centrifugal time, 24 min. And the best saturation of ammonium sulfate for enzyme purification was d5%, as the enzymatic activity and specific activity was 651.01 U and 34.099 U/g, respectively.
