摘要
建立了固相萃取-高效液相色谱(HPLC)测定番茄及番茄制品中番茄甙的方法。样品中的番茄甙经过甲醇提取,PCX固相萃取法净化,乙酰化后,采用高效液相色谱法进行测定。以乙腈和水(90:10)为流动相,经过C18色谱柱分离,紫外检测器检测。结果表明:番茄甙的检出限为0.5mg/kg,方法回收率为90.5%~93.5%,相对标准差为1.7%~3.2%。该方法准确、灵敏、适用于番茄及番茄制品中番茄甙的测定。
A method was developed for determination of tomatine in tomato and its processed products by high performance liquid chromatography(HPLC).The tomatine in sample was extracted by methanol,cleaned up with PCX solid-phase extraction cartridge,then acetylated by acetic anhydride and determined by HPLC.The mobile phase consisted of 90% acetonitrile and 10% water.The tomatine was separated on C18 column and determined with UV detector at 205nm.The results showed:the detection limit of tomatine was 0.5mg/kg,the average recoveries were from 90.5% to 93.5%,and the relative standard deviations(RSDs)was from 1.7% to 3.2%.This method was accurate,sensitive and effective for determination of tomatine in tomato and its processed products.
