摘要
目的初步探讨西宁地区藏族人群中HBV的流行及其基因型,为制定肝炎防治策略提供参考。方法应用ELISA检测HBV ayw型,PCR扩增s基因、c基因片断后测序,使用MEGA软件进行核苷酸序列的同源性分析并绘制进化树。结果ELISA只检测到7例ayw亚型,25例阳性的核苷酸序列测定,HBVS基因序列绝大部分聚集于系统发生树的D基因,样本中HBV的c基因序列则聚集于系统发生树中的基因型C。结论西宁地区藏族人群HBV基因绝大多数为C/D重组现象。
Objective To explore the prevalence and genotype of HBV of Tibetans in the Xining area, which would provide the strategy for hepatitis prevention and control. Methods HBV ayw was detected by ELISA method and S gene,C gene fragment were amplificated by PCR method and sequenced, and then the nucleotide sequence homology analysis and phylogenetic tree made by MEGA3 software. Results It was shown that 8 cases of ayw subtyp was detected by ELISA,25 cases were positive by nucleotide sequence determination of HBV S gene sequences, mostly gathered in the D gene of the phylogenetic tree,in the HBV sample of C gene sequences were gathered in the phylogenetic tree of genotype C. Conclusion HBV gene is C/D recombination mostly in population of Tibetans in the Xining area.
