摘要
建立湖羊促性腺激素释放激素受体(gonadotropin releasing hormone receptor,GnRHR)基因的实时荧光定量聚合酶链反应(RT-PCR)检测方法,并采用该方法对湖羊消化系统中胃肠各组织的GnRHR mRNA表达水平进行检测。结果表明:GnRHR mRNA在瘤胃、网胃、瓣胃、皱胃、十二指肠、空肠、回肠、盲肠、结肠和直肠部位均有表达,其中瘤胃GnRHR mRNA表达量最高,极显著高于其他各组织(P<0.01);网胃GnRHR mRNA表达量极显著高于十二指肠(P<0.01),显著高于瓣胃、盲肠和直肠(P<0.05);皱胃GnRHR mRNA表达量极显著高于瓣胃、空肠、回肠和盲肠(P<0.01),显著高于结肠和直肠(P<0.05);结肠GnRHR mRNA表达量显著高于十二指肠和直肠(P<0.05)。研究证实了GnRHR mRNA在湖羊消化系统中有表达,这为GnRH功能的多样性研究提供了新的依据。
To detect the gonadotropin releasing hormone receptor (GnRHR) mRNA expression pattern in sheep's digestive system, the SYBR green real-time fluorescence quantitative polymerase chain reaction (RT-PCR)method was established. With this method, the GnRHR mRNA expression in stomach and intestinum system of Hu sheep was detected. The results showed that GnRHR mRNA expressions were detected in rumen, reticulum, omasum, abomasums, duodenum, jejunum, ileum, caecum, colon and rectum. The G.nRHR mRNA expression in rumen was significantly higher than those in other tissues (P 〈 0.01) ; the expression in reticulum was significantly higher than that in duodenum (P〈 0.01) and higher than those in omasum, caecum and rectum (P 〈 0. 05); the expression in abomasums was significantly higher than those in omasum, jejunum, ileum and caecum (P 〈 0. 01), and higher than those in colon, rectum (P 〈 0.05) ; the expression in colon was higher than those in duodenum and rectum (P 〈 0.05). It is demonstrated that there are CmRHR mRNA expressions in the digestive system of Hu sheep. It provides new evidenees for the research field of GnRH functional diversity.
