高效液相色谱法测定人血清中拉莫三嗪浓度 被引量:8

Determination of lamotrigine in serum by HPLC

摘要 目的评价高效液相色谱测定人血清中拉莫三嗪浓度的方法。方法用Hypersil C18色谱柱,柱温为30℃,流动相为1%三(羟甲基)氨基甲烷-乙腈(70∶30),流速为0.8 mL.min-1,测定波长为230 nm。结果拉莫三嗪血清样品线性范围为0.39~50.00 mg.L-1,1.56,6.25,25.00 mg.L-13个浓度质控样品日内、日间精密度(RSD)均<5%,方法学回收率分别为(103.8±3.6)%,(100.0±2.2)%,(99.1±4.2)%。结论本方法回收率高、准确度好、操作简便,适用于临床上对拉莫三嗪血药浓度的常规监测。 Objective To establish a HPLC method of determination of lamotrigine concentration in serum.Methods Hypersil-C18 column was used,temperature of colum was 30 ℃,mobile phase was 1% Tris(hydroxymethyl) aminomethane-acetonitrile(70∶30),flow rate was 0.8 mL·min-1.The detecting wavelength was at 230 nm.Results The linear range of lamotrigine serum samples was 0.39%-50.00 mg·L-1.The intra-day and inter-day precisions of 3 different concentrations(1.56,6.25,25.00 mg· L-1) were all less than 5%.The methodology recoveries were(103.8± 3.6)%,(100.0±2.2)%,(99.1±4.2)%,respectively.Conclusion The method with high recovery,good accuracy,easy operation is suitable for clinical routine monitoring for serum concentrations of lamotrigine.
