摘要
[目的]探讨灵芝原生质体的高效制备及诱变方法。[方法]用2.5%溶壁酶、0.5%蜗牛酶和0.5%纤维素酶组成的复合酶液裂解适龄灵芝菌丝体,制备原生质体,统计其再生频率,并用制备出的原生质体进行诱变试验。[结果]采用该制备方法获得了高质量的原生质体,其再生率能稳定达到80%以上,且经复合诱变后发生基因突变的频率也较高。[结论]该研究为后续的灵芝诱变选育奠定了基础。
[Objective] The aim was to discussed a method of protoplast preparation and mutagenesis of Ganoderma lucidum.[Method] Use composite enzyme containing 2.5% lywallzyme,0.5% glusulase and 0.5% cellulase to split proper age Ganoderma lucidum mycelium,and after protoplast preparation,its regeneration rate was figured out.Then the mutagenesis test was done using the prepared protoplast.[Result] High-quailty protoplasts were obtained by this study’s preparation method and the regeneration rate could reach to over 80% stably.The mutation rate by mixed mutagenesis was also relatively high.[Conclusion] It prepared the ground for the following Ganoderma lucidum mutation breeding.
