三氧化二砷对大鼠成骨细胞增殖及BMP-2的影响

摘要 [目的]观察不同剂量三氧化二砷(As2O3)对体外培养的大鼠成骨细胞增殖、骨形成蛋白-2(BMP-2)基因表达的影响。[方法]不同浓度(10、1、0.1μmol/L)的三氧化二砷作用于体外培养的成骨细胞48和72 h后,观察成骨细胞的形态,采用MTT法检测成骨细胞的增殖,并通过RT-PCR检测成骨细胞BMP-2基因的表达情况。[结果]10μmol/L As2O3组可见部分成骨细胞变圆变小,细胞核固缩、碎裂,细胞膜皱褶、凹陷;1μmol/L As2O3组成骨细胞数目众多和分裂相明显;0.1μmol/L As2O3组成骨细胞间连接紧密。10μmol/LAs2O3组大鼠成骨细胞OD值明显低于对照组,而72 h OD值低于48 h OD值,有显著性差异(P<0.01);1μmol/L组As2O3OD值明显高于对照组,而72 h OD值高于48 h OD值,有显著性差异(P<0.01);0.1μmol/L As2O2组72 h OD值与48 h OD值无显著差异(P>0.05),且OD值与对照组无显著差异(P>0.05)。RT-PCR结果表明,10μmol/L组72 h BMP-2基因表达明显低于48 h,且BMP-2的表达明显低于对照组,有显著性差异(P<0.01);1μmol/L As2O3组72 h BMP-2基因表达与48 h无显著差异(P>0.05),且BMP-2基因表达与对照组(P>0.05);0.1μmol/L As2O3组72 h BMP-2基因表达明显高于48 h,且BMP-2的表达明显高于对照组,有显著性差异(P<0.01)。[结论]As2O3对体外培养大鼠成骨细胞增殖和BMP-2 mRNA表达具有双向调节作用,并且呈剂量-时间依赖性。 [ Objective ] To study the effects of different doses of arsenic trioxide on the proliferation and bone morphogenetic protein -2 ( BMP- 2) gene expression of cultured rat osteoblasts cells. [ Method] After the osteoblasts were treated by different concentrations of As2O3 (10, 1 and 0.1 μmol/L) for 48 h and 72 h, the morphology of osteoblasts was observed under inverted mieroscophy, the proliferation of osteoblasts was tested by MTU assay, and the BMP-2 gene expression of osteoblasts was determined with RT-PCR. [ Result] In 10 μmoL/L arsenic triox- ide treatment group, some osteoblasts become rounder and smaller, the nuclear was condensed and fragmented, and the cell membrane folded and depressed; in 1 μmol/L arsenic trioxide treatment group, there were large number of osteoblasts with obvious split phase; in O. 1 μmol/L arsenic trioxide treatment group, the osteoblasts were closely connected. The OD value of 10 μmol/L group was significantly lower than the control group, OD value in 72 h was lower than 48 h, which had statistical difference ( P 〈 0.01 ) ; OD value of 1 μmol/L group was significant- ly higher than the control group. OD value in 72 h was higher than OD value in 48 h,whieh had statistical difference(P 〈0. O1 ). The OD val- ue of 0.1 μmol/L group in 72 h had no statistical difference with 48 h(P 〉0.05 ) and was the same as control group. The BMP-2 gene expres- sion of 10 μmol/L group in 72 g was significantly lower than in 48 g and BMP-2 gene expression of 10 txmoL/L group was significantly lower than the control group, which had statistical difference(P 〈 0.01 ) ; BMP-2 gene expression of 1 μmoL/L group had no significant difference between in 48 h and in 72 h and the same as control group( P 〉 0.05 ). BMP-2 gene expression of 0.1 μmol/L group in 72 h was significantly higher than in 48 h and BMP-2 gene expression of 10 μmol/L group was significantly higher than the control group, which had statistical differ- ence( P 〈 0.01 ). [ Conclusion] As2O3 has a positive-negative regulatory role for cell proliferation and BMP-2 mRNA expression to rat osteo- blast in vitro, and presents a dose- time dependence.
