摘要
以水溶性生物高分子牛血清白蛋白(BSA)为载体,与小分子2,4-二羟基水杨醛缩甘氨酸希夫碱金属配合物(SalGlyM,M=Cu、Zn)结合,制得了水溶性生物高分子金属配合物结合体(SalGlyM@BSA)。采用FT-IR、UV-Vis、CD和SDS-PAGE电泳对其结构进行了表征。采用NBT光化学还原法考察了SalGlyM@BSA的抗O2.-活性,探讨了金属配合物结合体清除O2.-的机理。结果表明氨基酸希夫碱具有一定的抗O2.-活性,结合生物高分子载体后,SalGlyM@BSA具有良好的水溶性和O2.-清除能力。
The water-soluble biopolymer/metal conjugates ( SalGlyM@ BSA) were prepared by the small molecule metal complexes, 4-hydroxy-salieylaldehyde Glyeine Schiff-base metal complexes ( SalGlyM, M = Cu Zn, conjugating with water-soluble biopolymer, bovine serum albumin(BSA). The structure of SalGlyM@ BSA were characterized by FT-IR spectra, UV-Vis spectra, circular dichroism(CD) and SDS-PAGE electrophoresis. The superexide anion radical ( O2.-) scavenging activity of SalGlyM, BSA and SalGlyM@ BSA complexes were investigated by NBT photochemical reduction and the effect mechanism of scavenging O2.- was also explored. The results showed that Glycine Schiff-base complexes had a certain antioxidant activity. After combining SalGlyM into BSA,the biopolymer/metal conjugates (SalGlyM@ BSA)showed water solubility and excellent O2.- scavenging activity.
