摘要
利用胰蛋白酶、胃蛋白酶和碱性蛋白酶分别水解大豆分离蛋白,研究水解物对嗜热链球菌增殖的促进作用。以M17培养基为基础培养基,分别添加3种蛋白水解物作为实验组,以添加大豆分离蛋白作为对照组,培养嗜热链球菌,测定对其产酸和活菌数的影响;并通过蛋白层析技术对3种水解物进行组分分析。结果显示,碱性蛋白酶水解物对嗜热链球菌增殖的促进效果最为显著,使活菌数提高了1个数量级,菌株产酸能力增强。层析结果显示,大豆蛋白碱性蛋白酶水解物(AS)中短肽含量最高,是嗜热链球菌培养的一种理想氮源。
Trypsin, pepsin and alkaline protease are used for hydrolyzing soybean peotein isolate to test their effects on promoting the growth of Streptococcus thermophilus (S.t). M17 medium is used as basic medium, three kinds protein hydrolysates are added as experimental group, control medium is M17 medium with soybean protein, all of them are used to culture S.t and compare their effects on promotion of S.t, protein hydrolysates are analyzed by protein chromatography. The result shows that hydrolysates by alkaline protease can obviously increase the number of viable cells of S.t as one magnitude and content of peptide is high, which can provide a new optimum nutrigeon source for high-density cultivation of S.t.
