桃金娘茎枝中没食子酸的分离鉴定及含量测定 被引量:3

摘要 目的:对桃金娘茎枝中的没食子酸进行提取分离鉴定并测定含量。方法:采用80%乙醇对桃金娘茎枝进行提取,利用柱层析和重结晶对提取液进行分离,通过理化性质鉴别和波谱技术鉴定其结构;采用高效液相色谱法测定桃金娘茎枝中没食子酸含量,色谱柱:LichrospherC18,流动相:甲醇-0.2%磷酸水溶液(1.5∶98.5),流速1.0mL/min,检测波长为270nm,柱温为室温,进样量为10μL。结果:没食子酸在0.1~0.6μg范围内呈良好线性关系(r=0.9992),平均回收率为100.6%,RSD为1.32%(n=6)。结论:该方法简便可行,重现性好,可用于桃金娘茎枝中没食子酸含量测定。 Objective: To isolate and determine gallic acid in stem and branch of Rhodomyrtus tornentosa(Ait.)Has- sk.. Methods: Gallic acid was extracted by 80% ethanol, isolated and purified by column chromatography, and recrys- tallized from the stem and branch of Rhodomyrtus tornentosa(Ait. ) Hassk.. A HPLC method was set up to determine the content of gallic acid in Rhodornyrtus tornentosa (Ait.)Hassk., using Lichrospher C18 (4.6 mm× 250 mm, 5 μm) column, and methanol-0.2%-phosphoric acid water solution (1.5:98.5) as the mobile phase, the UV detection wavelength was at 270 nm, with the flow rate of 1.0 mL/min at room temperature. Results: A good linear was obtained in the range of 0.1 - 0.6 I^g and the average recovery was 100.6% (RSD =1.32%, n = 6). Conclusion: The method is simple with a good reproducibility for the determination of gallic acid in Rhodomyrtus tornentosa(Ait. )Hassk.
