摘要
目的:研究调更汤改善去势SD大鼠下丘脑线粒体氧化损伤的作用。方法:去势3月龄SD大鼠随机分为中药组(调更汤)、西药组(倍美力)、模型组,并设立未去势3月龄大鼠为青年组,实验8周。比较各组大鼠下丘脑E2、FSH、MDA、SOD、T-AOC、LDH含量,Mt-DNA损伤基因的mRNA表达,下丘脑神经元细胞线粒体微观结构变化。结果:①与青年组比较,模型组大鼠下丘脑E2水平显著降低(P<0.01),FSH水平显著升高(P<0.01);中药组大鼠下丘脑E2、FSH水平与模型组无显著差异(P>0.05);西药组大鼠下丘脑E2水平较模型组升高(P<0.01),FSH降低水平(P<0.01)。②模型组大鼠下丘脑MDA、LDH含量均较青年组显著升高(P<0.01),SOD、T-AOC水平显著降低(P<0.01);中药组和西药组MDA、LDH含量显著降低(P<0.01),SOD、T-AOC水平显著提高(P<0.05)。③模型组大鼠下丘脑线粒体损伤相关基因CoImRNA、CoIII mRNA的表达显著升高(P<0.01);中药组和西药组CoI mRNA、CoIII mRNA表达降低(P<0.01)。④中药组和西药组大鼠下丘脑线粒体微观结构均较模型组改善。结论:调更汤具有改善下丘脑线粒体氧化损伤作用。
Objective: To study the improvement effect of "Tiaogeng Decoction" for mitoehondrial oxidative damage of hypothalamus in SD rats. Methods: Castration rats of 3 months were randomized into Chinese medicine group, western medicine group and model group, and the rats with sex ability were enrolled into young group, with the course of 8 weeks. Compare the contents of E2, FSH, MDA, SOD, T-AOC, LDH, expression of Mt-DNA and microstructure changes of hypothalamic neuron mitoehondria. Results: (1) Compared with the young group, the level of E2 in model group was decreased significantly ( P 〈 0.01 ) , and level of FSH was increased significantly ( P 〈 0.01 ) ; there was no significant difference in the levels of E2 and FSH between Chinese medicine group and model group ( P 〉 0.05 ) ; the E2 level in western medicine group was higher than that in model group (P 〈0.01 ) , and the FSH level in western medicine group was lowered (P 〈0.01 ). (2) The contents of MDA and LDH in model group were higher than those in young group significantly ( P 〈 0.01 ) , and the levels of SOD and T-AOC were decreased significantly (P 〈 0.01 ) ; the contents of MDA and LDH in Chinese medicine group and western medicine group were decreased markedly (P 〈 0.01 ), the levels of SOD and T-AOC were increased markedly (P 〈 0.05). (3) Expressions of Col mRNA and Colll mRNA in model group were increased significantly (P 〈 0. 01 ); the expressions of CoI mRNA and Colll mRNA in Chinese medicine group and western medicine group were decreased ( P 〈 0.01 ). (4) The improvement of mierostrncture changes of hypothalamic neuron mitochondria was better in Chinese medicine group and western medicine group than in the model group. Conclusion: "Tiaogeng Decoction" can improve mitochondrial oxidative damage of hypothalamus.
