摘要
目的:探讨电针对心肌缺血再灌注损伤心肌的保护作用机制。方法:将50只大鼠随机分为5组:空白组,假手术组,模型组,高频电针组,低频电针组。5组动物经预处理5d后麻醉,全程记录心电图。空白组同期捆绑对照;假手术组开胸挂线不结扎;模型组、高频电针组和低频电针组开胸结扎大鼠左冠状动脉前降支,造成心肌缺血20min,再灌注40min。经以上处理后颈动脉取血,采用721分光光度计测定血清缺血修饰白蛋白(IMA),放射免疫法测定血浆降钙素基因相关肽(CGRP)。结果:模型组大鼠心肌缺血再灌注后心电图STⅡ值明显高于空白组、假手术组、低频针刺组和高频针刺组。模型组IMA、CGRP含量较空白组和假手术组明显升高,而低频电针组和高频电针组与之比较显著降低;高频电针组大鼠心肌缺血再灌注后的心电图STⅡ值、IMA和CGRP含量都低于低频电针组。结论:电针内关穴能降低心肌缺血再灌注损伤大鼠的STⅡ值,减少血液中IMA、CGRP含量。提示其抑制缺血再灌注应激反应的发生,参与缺血心肌的保护,其中高频组的作用优于低频组。
Objective:To discuss the mechanism of electro-acupuncture in protecting the myocardial ischemia reperfusion injured muscle.Methods:50 rats were randomly divided into five groups,including blank control group①,sham operated group②,model group③,lower frequency of electro-acupuncture group④ and higher frequency of electro-acupuncture group⑤.All these five groups were bound in anesthesia and their ECG were recorded during the whole process.The blank group was bound as the control at the same time.The sham operated group was operated to open the thoracic cavity and was braided through the artery with no ligation;the latter three groups must be operated to open their thoracic cavity and ligate left descending coronary artery,to result in myocardial ischemia for 20 minutes,and reperfusion for 40 minutes following.After all manipulation above,we took carotid artery blood to measure IMA by spectrophotometer and CGRP by radio-immunity method.Results:The value of STⅡ in ECG of model group after MI and reperfusion was significantly higher than those in the blank group and sham operated group,moreover higher than those in high frequency manipultion group and low frequency manipulation group.The IMA and CGRP content of model group were increased significantly,compared with ① and ②,but decreased in ④ and ⑤.The value of STⅡ in ECG,IMA and CGRP of ④ were lower than ⑤ after MIRI.Conclusion:Electro-acupuncture at Neiguan point can decrease the IMA,CGRP content in rats blood with myocardial ischemia and reperfusion and reduce the value of STⅡ in ECG.It restrains the stress reaction to protect the heart muscles.Meanwhile,the acupuncture effect of the high frequency manipulation group is superior to that of the low frequency manipulation.
