摘要
目的观察妊娠前后补充叶酸对妊娠期邻苯二甲酸二-(2-乙基己基)酯(DEHP)暴露致孕鼠及其胚胎损伤的防治作用。方法将50只雌鼠随机分为5组,空白组给予生理盐水,玉米油组和DEHP组给予玉米油,叶酸组给予叶酸60 mg/(kg.d),实验组给予玉米油+叶酸。7 d后制备孕鼠模型,妊娠后大鼠继续妊娠前给药,DEHP组及实验组另添加DEHP,均灌胃,剂量同。于妊娠19 d处死孕鼠取胚,记录孕鼠体质量、肝脏、子宫、卵巢质量,统计胚胎植入总数、吸收胎数、活胎数、死胎数,观察胚胎外形,测量并计录胎鼠体质量及胎盘质量。结果实验组孕鼠肝脏、子宫、卵巢指数分别为4.35±0.88、1.27±0.11、0.088±0.013,与DEHP组相比具有显著性差异(P均<0.05),与余3组相比,P均>0.05;实验组活胎数、胎鼠体质量及胎盘质量分别为(12.34±2.19)个、(5.78±0.53)g、(1.04±0.10)g,与DEHP组相比,P均<0.05,与余3组相比,P均>0.05。结论妊娠前后补充叶酸可以降低妊娠期DEHP暴露对孕鼠及其胚胎造成的毒性损伤作用。
Objective To observe the protective effects of folic acid on embryotoxicity induced by di-(2- ethylexyl) (DEHP) phthalate in rats. Methods 50 female rats were randomized equally into five groups. The blank group was ad- ministered with 0.9% Sodium Chloride, the corn oil group and DEHP group with corn oil, the folic acid group with 60 rag/ (kg . d) folic acid, and the experimental group with corn oil and folie acid. The model of pregnant rats were established after 7 days. All the rats were administered with corresponding drugs everyday from the beginning of the experiment to the end. The DEHP and experimental group were administered with equal dose DEHP in addition after pregnancy. The preg- nant rats were killed and the fetal rats were removed on 19th day. The following resuhs in each group were recorded: the body weight and the organic weight of the female rats, number and weight of fetus rats and the placental weight. Results The coefficient of fliver, uterus and ovary/body in experimental group were 4.35 ± 0.88, 1.27 ± 0. 11 and 0. 088 ± 0.013, respectively, compared with DEHP group, all P 〈 0.05. Compared with other three groups, there was no statistical significance(all P 〉 0.05 ). In experimental group, the fetal quantity, fetal and placental weight were 12.34 ± 2. 19, ( 5.78 ± 0.53 ) g and ( 1.04 ± 0.10) g, compared with DEHP group, all P 〈 0.05, compared with other three groups, there was no statistical significance(all P 〉 0.05). Conclusion Folic Acid supplementation before pregnancy can have a pro- tective effect to the female rats and fetal rats exposed in DEHP.
