摘要
目的通过检测实验性自身免疫性脑脊髓炎(EAE)大鼠中NLRP3信号传导通路的改变,探讨NL-RP3炎性小体介导的释放IL-1β的炎性反应在EAE大鼠发病机制中的作用。方法将30只大鼠随机分为两组,正常对照组10只,EAE模型组20只,并以豚鼠脊髓匀浆(GPSCH)为抗原免疫建立EAE模型。免疫组化检测比较对照组和模型组大鼠CNS内NLRP3、caspase-1、IL-1β细胞因子的表达情况并进行研究分析。结果模型组脑和脊髓的NLRP3、caspase-1、IL-1β的表达均高于正常对照组(P<0.05)。结论 NLRP3介导炎性反应可能参与了EAE的发病过程,可能是MS的发病机制之一。
Objective To detect changes of NLRP3 signal transduction pathway of experimental autoimmune encephalomyelitis(EAE) to analyze NLRP3-mediated IL-1β release inflammatory process in rats.Methods Rats were randomly divided into two groups,10 rats in the control group,20 rats in the model group.EAE model was induced in rats by immunization with guinea pig spinal cord homogenate(GPSCH).The expression of NLRP3,caspase-1 and IL-1β in the CNS of two groups was detected by immunohistochemistry.Results The expression of NLRP3,caspase-1 and IL-1β in the model group was more than the control group in every part(P0.05).Conclusion NLRP3-mediated inflammatory response probably involved in the process of the EAE,so it maybe one of the pathogenesis of multiple sclerosis.
