黄芪多糖对大鼠急性颈脊髓损伤后血清及肺泡灌洗液CD44含量变化的影响 被引量:6

Role of astragalin to CD44 in serum and bronchoalveolar lavage fluid in pulmonary edema in acute cervical spinal cord injured rats

摘要 [目的]探讨大鼠急性颈脊髓损伤后黄芪多糖与CD44含量变化的关系。[方法]成年Wistar大鼠80只,体重240~250 g,雌雄不限。大鼠随机分为四组,分别为黄芪多糖组、生理盐水组、空白对照组、假手术组,每组20只。各组按取材时间不同又分为造模后24 h、3 d、1周、2周共4个时间点,每个时间点5只大鼠。采用颈7段脊髓改良Allen's打击法制作大鼠脊髓损伤模型,打击力度为10×2.5g.cm;假手术组仅暴露颈7段脊髓。[结果]在不同时间点处死大鼠,测量各时间点每组大鼠血清和肺泡灌洗液中CD44含量。结果显示黄芪多糖对脊髓损伤大鼠CD44含量变化有影响。伤后3d黄芪多糖组血清及肺泡灌洗液CD44含量较空白对照组和生理盐水组有显著差异性(P<0.05)。[结论]黄芪多糖可以减轻大鼠急性颈脊髓损伤后血清CD44下降幅度并提高伤后3d肺泡灌洗液CD44含量。这一结果可能对治疗急性脊髓损伤后肺水肿有帮助。 [ Objective ] To investigate the relationship between astragalin and the deviation of CD44 at different time points in acute cervical spinal cord injured rats. [ Method] Eighty wistar rats with 240 g to 250 g in weight were randomly divided into four groups:astragalin group, normal saline group, blank group and sham group, which were subdivided into groups of 24 hours, 3 days, 1 week and 2 weeks after injury respectively. The spinal cord injury (SCI) models were induced at the level of seven cervical vertebrae (C7 ) by dropping a weight of 10 g from the height of 25 mm (Allen~ model). In the sham group, laminas were removed only, leaving spinal cord at C7 intact. [ Result ] All the animals were sacrificed at time points ranging from 24 hours to 2 weeks (24 h, 3 d, 1 week, 2 weeks) after injury. The content of CD44 in serum and bronchoalveolar lavage fluid (BALF) was harvested. There was statistical significance to the content of CD44 between astragalin group and normal saline group and blank group at 3d after injury(P 〈 0.05). [ Conclusion] Astragalin can decrease the decline of CD44 in serum and increase the content of CD44 in BALF at 3d after injury in acute cervical spinal cord injured rats. This may be help to the treatment of pulmonary edema in SCI patients.
