摘要
目的:研究重铬酸钾对大鼠睾丸支持细胞中雄激素结合蛋白(ABP)、转铁蛋白(Tf)和抑制素(INH)蛋白的表达的影响。方法:建立大鼠睾丸支持细胞体外原代培养模型,分为0mol/L、10-7mol/L、10-6mol/L重铬酸钾染毒组,Westernblot法检测重铬酸钾作用24h后各组睾丸支持细胞中ABP、Tf和INH蛋白表达水平的变化。结果:ABP蛋白的表达量随重铬酸钾浓度的升高而升高(0.39±0.19、0.51±0.18及0.81±0.19),差异有统计学意义(F=10.738,P<0.001);Tf蛋白的表达量也随重铬酸钾浓度的升高而增加(0.56±0.19、1.21±0.19及1.19±0.17),差异有统计学意义(F=32.435,P<0.001);而INH蛋白表达却呈下降趋势(1.06±0.19、1.20±0.19及0.71±0.19),差异有统计学意义(F=14.116,P<0.001)。结论:重铬酸钾能够引起大鼠睾丸支持细胞分泌功能的变化,产生生殖毒性作用。
Aim:To observe the effect of potassium bichromate on the expressions of ABP,Tf and INH protein in rat sertoli cells(SC).Methods:Rat SC were isolated and cultured with 0 mol/L,10-7 mol/L,10-6 mol/L potassium bichromate for 24 h.Western blot was used to determine the expressions of ABP,Tf and INH protein.Results:With the increasing concentration of potassium bichromate,the expression level of ABP(0.39 ± 0.19,0.51 ± 0.18,and 0.81 ± 0.19;F = 10.738,P 〈 0.001) and Tf protein(0.56 ± 0.19,1.21 ± 0.19,and 1.19 ± 0.17;F = 32.435,P 〈 0.001) increased;while that of INH protein decreased(1.06 ± 0.19,1.20 ± 0.19,and 0.71 ± 0.19;F = 14.116,P 〈 0.001).Conclusion:Potassium bichromate can affect the secretory functions of rat SC,and produce reproductive toxicity.
