摘要
目的建立HPLC法测定雪莲种子提取物中牛蒡子苷含量的方法,为新疆雪莲特征性化合物的指认提供科学依据。方法采用Phenomenex Gemini色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水梯度洗脱,流速1.0 ml/min,柱温30℃,检测波长270 nm。结果牛蒡子苷含量为0.24%且与其他成分分离良好,在105~1 050μg/ml范围内线性关系良好,r=0.999 7(n=6),对照品溶液低、中、高三种浓度的回收率分别为95.88%,102.33%,107.05%;RSD分别为3.88%,1.38%,1.02%。结论所建立的方法简便、准确、重现性好,并具有良好稳定性,为准确测定雪莲种子中牛蒡子苷含量提供可靠依据。
Objective To establish an HPLC method for the determination of arctiin in seeds of Saussurea involucrata,and to provide the scientific basis for the identification of characteristic compound of Saussurea involucrata.Methods Use Phenomenex Gemini column(250 × 4.6 mm,5 μm);gradient elution of methanol and water with a flow rate of 1 ml / min.Column temperature was controlled at 30℃ while the detection wavelength was 270 nm.Results With a good resolution to other constituents,the arctiin content was 0.24%.The linear range was 1.05 ~ 1050 μg / ml,r = 0.9997.In three concentration levels(low,middle,high),recovery rates of control solution were 95.88%,102.33%,107.05%,with the RSD of 3.88%,1.38%,1.02%,respectively.Conclusion This method is simple,accurate,and has good stability,while provide a reliable basis for the accurate determination of arctiin in seeds of Saussurea involucrata.
