摘要
【目的】探讨自噬在血管紧张素(Ang)Ⅱ诱导心肌细胞肥大中的作用。【方法】原代培养的乳鼠心肌细胞,采用AngⅡ干预建立心肌肥大的模型;在光学显微镜下观察心肌细胞的形态,采用Real time PCR技术检测心肌细胞肥大标志物心房利钠多肽(ANP)的表达、Western blot法检测LC3蛋白的表达。【结果】AngⅡ可抑制自噬相关基因LC3蛋白的表达。自噬阻断剂3-甲基腺嘌呤(3MA)会促进AngⅡ诱导的心肌细胞肥大。【结论】AngⅡ可能通过抑制自噬来诱导心肌细胞肥大。
[ Objective ] To investigate the role of autophagy in cardiac hypertrophy induced by angiotensin Ⅱ (Ang Ⅱ ). [ Methods ] Cardiac hypertrophy was induced by Ang Ⅱ incuhured neonatal rat cardiomyocytes. Morphology and expression of hypertrophy marker atrial natriuretic peptide (ANP) were detected by optical microscope and real time-PCR. Autophagic marker, LC3 level was tested by Western blotting. [ Results ] AngⅡ increased volume of cardiomyocyte, and up regulated RNA expression of ANP. Ang Ⅱ inhibited autophagy related gene LC3 protein expression. Blocking autophagy by 3-MA could significantly enhance myocardial hypertrophy induced by Ang Ⅱ. [ Conclusion] Ang Ⅱ may induce myocardial hypertrophy by inhibiting autophagy.
