摘要
目的:利用反相高效液相色谱法研究氨茶碱在家兔体内的药代动力学。方法:色谱柱为HypersilBDSC18色谱柱(4.6×200mm,5μm),流动相为甲醇:水(25:75),检测波长274nm。结果:家兔静脉注射氨茶碱后,主要药代动力学参数为:Tmax=(0.17±0)h,Cmax=(87.068±6.17)μg/ml,t1/2=(4.658±1.587)h,AUC0-∞=(304.931±48.661)μg·h/ml,MRT=(5.877±1.215)h,CL=(0.039±0.006)L/h/kg。结论:本方法简便,准确,灵敏度高,重现性好,能够满足氨茶碱在家兔体内的药代动力学研究,为人体内氨茶碱的药代动力学研究提供参考。
Objective to study the pharmacokinetic process about the concentration in rabbits plasma of Aminophylline with RP-HPLC method.Methods RP-HPLC analysis was carried out by using Hypersil BDS C18 column (4.6×200mm,5μm) and methanol-water (25:75) as the mobile, and the UV detection wavelength was set at 274nm. Results The pharmacokinetic parameters of Aminophylline in rabbits were as follows: Tmax=(0. 17±0)h,Cmax=(87.068±6.17)μg/ml, t1/2=(4.658±1.587)h, AUC0-∞= (304.931±48.661)μg·h/ml, MRT=(5.877±1.215)h,CL=(0.039±0.006)L/h/kg. Con- clusion of the method was simple, accurate, highly sensitive and reproducible. It was a good reference for pharmacokinetics in rabbits and further for human bodies.
