摘要
目的探讨丹参注射液对小鼠急性酒精性肝损伤保护作用的机制。方法 60只小鼠随机分为正常对照组、模型组、丹参注射液低剂量组(10g.kg﹣1.d﹣1)、丹参注射液高剂量组(20g.kg﹣1.d﹣1),连续腹腔注射10d后,除正常对照组外,其余均灌胃给予50%乙醇12ml.kg﹣1制作小鼠急性酒精性肝损伤模型。禁食不进水8h后,检测肝指数,肝功能指标谷丙转氨酶(ALT)、谷草转氨酶(AST)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,并行肝脏病理学检查。结果丹参注射液两个剂量组可以明显降低急性酒精性肝损伤模型小鼠肝指数、血清ALT、AST活性和肝组织MDA含量,升高肝组织中GSH-PX活性,改善小鼠肝脏病理形态学改变。结论丹参注射液对急性酒精性肝损伤小鼠具有一定保护作用,其机制可能与抗脂质过氧化损伤有关。
Objective To study the protective effects of Salvia miltiorrhiza injection on acute alcohol induced liver injury in mice. Methods Sixty mice were randomly divided into four groups,The normal control group,the model grouP,low-dose(10g·kg^-1·d^-1) of Salvia miltiorrhiza injection and high-dose(20g·kg^-1·d^-1) of Salvia miltiorrhiza injection were given daily intraperitoneally for a period of 10 days,The control grouPand model grouPwere given an equal volume of saline.Seven days later,in addition to the normal group,the remaining groups were given by oral administration of 50% ethanol 12 ml·kg^-1 to replicate acute alcohol induced liver injury model.After absolute diet and no water fasting for 8 hours,the liver index,the alanine aminot ransferase(ALT) and aspartateaminotransferase(AST) in serum,and malondialdehyde(MDA) content and glutathione peroxidase(GSH-Px) activity in liver tissue were detected.The histological changes in liver were evaluated by hematoxylin-eosin(HE) staining.Results In the low-and high-dose drug treatment groups,Salvia miltiorrhiza injection significantly reduced liver index,the serum levels of ALT,AST and the contend of MDA,and elevated the activity of GSH-Px.In addition,Salvia miltiorrhiza injection could obviously improve pathological changes of liver tissue.Conclusion Salvia miltiorrhiza injection have some protective effects for acute alcoholic liver injury in mice.The mechanism may be related to Resisting to the Lipid Peroxidation.
