三维质谱成像技术在研究脑神经肽和磷脂空间分布中的应用 被引量:1

摘要 目的利用三维质谱成像技术(mass spectrometry imaging, MSI )检测黄道蟹脑组织中多种神经肽家族和脂类的分布情况。方法通过结合组织直接进样和以基质辅助激光解吸电离串联飞行时间质谱(matrix assisted laser desorption ionization-time of flight/time of flight mass spectrometry, MALDI-TOF/TOF MS )为基础的成像技术以及形态学相关信息,在蟹脑组织中检测并分析神经肽和脂类的分布情况。结果通过神经肽直接组织分析检测到速激肽相关肽1a、RFamide族神经肽、orcokinin家族三种神经肽图谱,不同的神经肽家族具有不同的定位模式。共榆测到来源于8个家族的28种神经肽,所有神经肽主要定位在神经脑叶附近。脂类的鉴定结果显示卵磷脂平均分布于脑的各个区域,鞘磷脂在神经纤维中丰度较高。结论通过MALDI—TOF/TOFMS方法,并结合MSI技术,获得了黄道蟹脑组织中多种神经肽和脂类的三维图像,二者在脑组织中的三维分布模式和空间定位表明其在神经系统中可能具有不同的功能。 Objective To examine distribution patterns of multiple neuropeptide families and lipids in the brain of the partan by a three-dimensional mass spectrometry imaging(3D MSI) technique. Methods The distribution of numerous neuropeptides and lipids in carb brain tissue were detected and analyzed by the matrix assisted laser desorption ionization-time of flight/time of flight mass spectrometry (MALDI-TOF/TOF MS) combined with direct probe inlet and morphological information. Results There were three neuropeptides spectrum of a graph including CabTRPla m/z934.5, RFamide tribe neuropeptide and orcokinin tribe neu- ropeptide were detected by direct tissue analysis. Different localization patterns were observed for different neu- ropeptide families. There were 28 kinds neuropeptides from 8 families were found,and all the neuropeptides were located nearby lobe of brain. Lipidic assessment results showed that phosphotidylcholine distributed me- dially each area of brain tissue,sphingomyeline had higher abundance in nerve fibers. Conclusion The 3D structure of numerous neuropeptides and lipids in partan brain tissue are determined by MSI combined with MALDI-TOF/TOF MS, their different localization patterns suggesting their potential different functional roles in the nervous system.
