祖师麻总香豆素的理化性质研究 被引量:1

摘要 目的考察祖师麻总香豆素的平衡溶解度和油水分配系数等理化常数,及其吸湿性和稳定性。方法采用紫外分光光度法,测定祖师麻总香豆素在不同介质中的平衡溶解度、不同pH条件下的油水分配系数(Papp)、不同湿度条件下平衡吸湿量;以祖师麻总香豆素含量变化率为指标,考察光照、高温和高湿条件对其稳定性的影响。结果祖师麻总香豆素在水、0.1mol.L-1HCL、pH 6.86磷酸缓冲液中的平衡溶解度(48h)分别为1.83mg.mL-1、2.07mg.mL-1、1.17mg.mL-1;祖师麻总香豆素在pH 1.2,2.1,3.5,4.7,5.8,7.4,8.9下的PapP值分别为12.14,11.67,10.98,10.11,9.86,9.12,8.63;祖师麻总香豆素在相对湿度为23%、43%、56%、59%、69%、75%、85%、95%条件下的含水量分别为10.10%、12.57%、15.37%、23.74%、32.98%、45.54%、56.90%、68.14%;祖师麻总香豆素在光照度为4500lx条件下5d和10d的含量变化率分别为99.6%和99.2%;在60℃条件下5d和10d的含量变化率为100.2%和98.5%,在相对湿度为95%的室温条件下5d和10d的含量变化率分别为90.6%和87.9%。结论祖师麻总香豆素的PapP值随着pH增大而减小,具有pH依赖性,表明其亲脂性较强;温度和光照对祖师麻总香豆素的含量和重量影响较小;祖师麻总香豆素的临界相对湿度为59%,在高湿度条件下吸湿明显,因此应在干燥条件下保存。 OBJECTIVE To study the physicochemical properties of total coumarin in cortex daphnes.METHODS The Physicochemical properties of total coumarin in cortex daphnes was characterized by experiment of balance solubilities,patition coefficient,balance curve of absorb and effection on stability.RESULTS The solubilities of Total coumarin extraction in water,acid,and orthophosphoric acid condition was 1.83mg·mL-1,2.07mg·mL-1 and 1.17mg·mL-1 after 48h;The Papp of it in pH 1.2,2.1,3.5,4.7,5.8,7.4,8.9 was 12.14,11.67,10.98,10.11,9.86,9.12,8.63.Water content of it in relative humidity of 23%,43%,56%,59%,69%,75%,85% and 95% was 10.10%,12.57%,15.37%,23.74%,32.98%,45.54%,56.90%,68.14%;The content changing rate of it in 4500lx were 99.6% and 99.2%,in the day of 5th and 10th.Hygroscopic changing rate was 90.6% and 87.9% in 5d and 10d.CONCLUSION The extraction′s Papp goes down with pH increasing,indicating its strong lipophicity.The relative moisture sorption of it is 59%;Temperature and light can′t affect its content and weight apparently.But its content and weight are chang a lot in humid condition,we need to keep it in dry condition.
