摘要
目的:建立同时测定桂枝挥发油中α-蒎烯、β-蒎烯和柠檬烯含量的方法。方法:采用气相色谱法。色谱柱为DB-17石英毛细管柱(30m×0.25mm×0.25μm),检测器为氢火焰离子化检测器(FID),采用程序升温(起始温度为60℃,以5℃.min-1升至110℃),流速为1mL.min-1,分流比为10:1,进样量为1μL。结果:α-蒎烯、β-蒎烯、柠檬烯的检测浓度分别在0.02285~0.22850mg.mL-1(r=0.9998)、0.00877~0.08770mg.mL-1(r=0.9996)、0.00448~0.04480mg.mL-1(r=0.9998)范围内同其与内标的峰面积比呈良好线性关系;三者平均加样回收率分别为95.9%、96.3%、96.9%,RSD分别为2.3%、2.0%、1.6%(n均为9)。结论:该方法简便、快速、准确,可用于同时测定桂枝中α-蒎烯、β-蒎烯、柠檬烯的含量。
OBJECTIVE:To establish a method for the simultaneous determination of α-pinene,β-pinene and limonene in volatile oil from Cinnamomi Ramulus.METHODS:GC method was adopted.The determination was performed on DB-17(30 m×0.25 mm×0.25 μm)column.The detector was FID;the initial temperature was 60 ℃,then programmed from 60 ℃ to 110 ℃ at 5 ℃.min-1 ;the flow rate was 1 mL.min-1 ;split injection was conducted with split radio of 10:1;injection volume was 1 μL.RESULTS:The linear ranges of α-pinene,β-pinene and limonene were 0.022 85~0.228 50 mg.mL-1(r=0.999 8),0.008 77~0.087 70 mg.mL-1(r=0.999 6),0.004 48~0.044 80 mg.mL-1(r=0.999 8),respectively;mean recoveries of the three components were 95.9%(RSD=2.3%),96.3%(RSD=2.0%),96.9%(RSD=1.6%),respectively(n=9).CONCLUSION:The method is simple,fast and accurate for the determination of α-pinene,β-pinene and limonene in Cinnamomi Ramulus.
