摘要
通过正交试验研究亮菌液体发酵的条件和菌丝体多糖的提取工艺,并对多糖采用DEAE-Cellulose-52进行分离。结果显示,亮菌液体发酵接种量为10%,转速为120 r/min,pH为5.5,培养时间7 d;水浴提取亮菌菌丝体多糖的最佳工艺是:料液比1︰60,水浴时间3 h,水浴温度80℃。经过DEAE-Cellulose-52层析柱获得一个多糖组分,紫外分析表明,这个组分在200 nm~400 nm都无紫外吸收。
Study on the conditions of liquid fermentation with Armillariella tabescens and extrac tmg polysacclaandes through orthogonal experiments. The experiment results showed that the optimum conditions of liquid fermentation are as follows: optimal seed inoculation size 10%, pH 5.5, the shaking speed 12 r/min and fermentation time 7 d. The optimum conditions for extracting polysaccharides from Armillariella tabescens mycelia are as follows: material- liquid ratio 1 : 60, temperature 80 ℃ and time 3 h. There are one component are obtained through DEAE -Cellulose-52 column chromatography. UV spectrophotometry revealed that the component ofArmillariella tabescens have no obvious absorption in 200 nm-400 nm.
