发酵乳酪乳清中生物活性肽对3T3-L1小鼠脂肪细胞PPARγ mRNA表达及NF-κB活性的影响 被引量:2

摘要 探讨新疆传统发酵乳酪乳清中生物活性肽(简称乳清活性肽)对炎症状态下3T3-1小鼠脂肪细胞PPARγmR-NA表达及NF-κB活性的影响,并探讨其可能的作用机制。体外培养小鼠胚胎成纤维细胞3T3-L1,经0.15 mmol/L 3-异丁基-1-甲基黄嘌呤、0.125μmol/L地塞米松和10 mg/L胰岛素诱导成为成熟的脂肪细胞后,给予脂多糖(LPS)刺激使脂肪细胞成炎症模型,再给予乳清活性肽(1,10,20,50 mg/L),以及阳性对照药高密度脂蛋白(10,50,100 mg/L),测定脂肪细胞中PPARγmRNA表达及NF-κB的活性。PPARγ在分化成熟的脂肪细胞有较高水平的表达,LPS诱导后表达下降,PPARγ的表达在HDL和乳清活性肽干预后明显升高,并且乳清活性肽20,50 mg/L组、HDL 50,100 mg/L组与LPS致炎组相比有统计学差异;NF-κB活性在LPS致炎后升高,与正常组相比有统计学意义(P<0.05),HDL和乳清活性肽干预组中NF-κB活性明显减低,并且乳清活性肽20,50 mg/L浓度组,HDL 50,100 mg/L组与LPS致炎组相比有统计学意义(P<0.05)。乳清活性肽可能是通过上调PPARγmRNA的表达以及降低NF-κB活性来发挥抗动脉粥样硬化的作用。 This study was to explore the effect of active peptides extracted and isolated from traditional Xinjiang fermented cheese whey( whey active peptide) on the expression of PPAR3, mRNA and activities of NF-κB in the inflammatory state of 3T3-L1 preadipocyte. 3T3-L1 were cultured and induced into maturation, then stimulated with LPS to induce the inflammatory state, given at different concentrations of whey active peptides (1, 10, 20, 50 mg/L), and positive control group HDL( 10, 50, 100 mg/L), then the expression of PPARγ mRNA, and the activity of NF-κB were measured after the cells were collected. PPAR3, mRNA were highly expressed in the matured 3T3-1 adipocyte stimulated with LPS. Compared with LPS group, the expression of PPARγ mRNA were increased significantly in 50, 100 mg/L HDL group and 20, 50 mg/L active peptide group. The activity of NF-κB were increased significantly in LPS group compared with the control group, while decreased significantly in 50, 100 mg/L HDL group and 20, 50 mg/L active group compared with LPS group. Whey active peptides can up- regulate the expression of PPARγ mRNA and decrease the NF-κB activity.
