摘要
内质网型ω-3脂肪酸去饱和酶(FAD3)是不饱和脂肪酸合成途径中催化十八碳二烯酸[18∶2(9,12)]转化为十八碳三烯酸[18∶3(9,12,15)]的关键酶。通过RT-PCR的方法从番茄叶片克隆到LeFAD3的全长cDNA,基因全长为1 184bp,开放阅读框(ORF)为1 134bp,注册号为EU251190。将该基因正反向序列分别插入带有35S-CaMV启动子pBI121表达载体下游,获得正、反义表达载体,转化农杆菌LBA4404,利用农杆菌介导法转化番茄,获得转基因番茄植株。
The endoplasmic reticulum omega-3 fatty acid desaturase(FAD3) is the key enzyme which catalyzes 18∶2(9,12) to form 18∶3(9,12,15) in the unsaturated fatty acid biosynthesis.The full length of the cDNA of LeFAD3 was isolated by RT-PCR from tomato leaves,which contains of 1 184 bp nucleotides and a 1 134 bp open reading frame(ORF) of 1 134 and accession number EU25119.The full-length sequences of sense-and antisense-LeFAD3 were inserted into pBI121 vector downstream of the 35S-CaMV promoter to form sense constructs and antisense constructs.The 35S-CaMV LeFAD3 constructs were first introduced into Agrobacterium tumefaciens LBA4404.Leaf disk infected with A.tumefaciens transformation was used to transform tomato.The transgenic plants of tomato were obtained.
