摘要
对苦参内生真菌BS003的液体发酵条件进行优化,以获得最大的发酵产物。在Plackett-Burman试验设计基础上,采用响应曲面法,以生物量为考察指标,优化发酵条件。结果表明,液体发酵最佳条件组成为:250mL锥形瓶中的培养基装液量为75mL、接种量104 cfu/mL、硝酸铵1.22g/L、马铃薯200g/L、蔗糖10g/L、乙酸钠1.66g/L、温度27℃、pH=7、转速150r/min、时间4d。回归分析表明,整个模型显著,此方法合理可行,相比优化前产量提高38%。
The endophytic fungi BS003 is separated from Sophora flavescens which have many medicinal values.The aim of this study is to optimize fermentation conditions of endophytic fungi BS003,and to obtain the maximal product of fermentation.The paper optimized fermentation conditions of endophytic fungi BS003 use response surface methodology on the basis of result of Plackett-Burman and the content of mycelium in liquid fermentation was used as index.The results showed that the optimum liquid fermentation conditions were as follows: 75 mL in 250 mL container,concentration of fungus 104 cfu/mL,NH4NO3 1.22 g/L,potato 200 g/L,sucrose 10 g/L,CH3COONa 1.66 g/L,temperature 27 ℃,initial pH 7.0,shaker speed 150 r/min,fermentation for 4 d.The whole model was significant and the method was reasonable.The product of fermentation increased 38% than before.
