IGF-1修饰的神经干细胞对阿尔茨海默病大鼠学习记忆能力的影响 被引量:4

摘要 目的观察胰岛素样生长因子-1(IGF-1)修饰的神经干细胞移植对阿尔茨海默病(AD)大鼠学习记忆能力的影响,并检测其在AD病大鼠脑内的存活及基因产物的表达。方法构建IGF-1逆转录病毒表达载体,转染到神经干细胞中,SD大鼠随机分为正常对照组、AD模型组、神经干细胞移植组、IGF-1修饰的神经干细胞移植组,除正常对照组外制备AD模型。移植4 w后,行Y迷宫检测大鼠的学习记忆能力;30 d后,处死实验鼠,通过HE染色观察各组实验鼠脑组织形态学改变。将BrdU标记的IGF-1基因修饰神经干细胞移植入穹窿海马伞切断的大鼠侧脑室。移植4 w后,用免疫组织化学方法检测IGF-1修饰的神经干细胞在脑内的基因表达情况。结果神经干细胞移植后AD大鼠的学习记忆能力明显改善,与AD模型组相比差异具有统计学意义(P<0.05)。其中IGF-1修饰的神经干细胞移植组表现最为明显,与正常对照组相比无明显差异(P>0.05),其形态学表现接近于正常对照组。结论 IGF-1修饰的神经干细胞移植后AD大鼠的学习记忆能力增强,脑组织病理改变减轻,学习记忆能力明显改善,作用明显优于神经干细胞移植组。
