肿瘤相关基因N—myc、Fas、MTA1、nm23-H1对子宫颈癌细胞增殖转移的影响 被引量:2

Influence of tumor related gene N-myc, Fas, MTA1 and nm23-H1 on cervical cancer cell proliferation and metastasis

摘要 目的探讨原癌基因N—myc、抑癌基因Fas、肿瘤转移促进基因MTA1、转移抑制基因nm23-H1对子宫颈癌细胞增殖转移的影响。方法采用实时荧光定量RT—PCR检测正常子宫颈组织30例,子宫颈浸润癌(均为子宫颈鳞状细胞癌)未转移组及转移组标本组织各30例中N—myc、Fas、MTA1、nm23-H1基因表达情况并进行比较分析。结果子宫颈浸润癌组织中N—myc、MTA1基因扩增水平均高于正常宫颈组织,差异有统计学意义(P〈0.05);子宫颈浸润癌组织中Fas、nm23-H1基因扩增水平均低于正常宫颈组织,差异有统计学意义(P〈0.05);子宫颈浸润癌组织转移组中MTA1基因扩增水平均高于未转移组,nm23-H1基因扩增水平均低于未转移组,差异均有统计学意义(P〈0.05);子宫颈浸润癌组织转移组与未转移组中N-myc、Fas基因扩增水平比较差异均有统计学意义(P〈0.05)。结论N-myc、MTA1基因促进子宫颈癌细胞增殖,Fas、nm23-H1基因抑制子宫颈癌细胞增殖。MTA1基因促进子宫颈癌细胞转移,nm23-H1基因抑制子宫颈癌细胞转移,且N—myc、Fas基因表达与子宫颈癌细胞转移亦有相关性。 Objective To investigate the influence of the original cancer gene N-myc, tumorsuppressor gene Fas, tumor metastasis gene MTA1 and metastasis-suppressor gene nm23-H1 on cervical cancer. Methods By real time fluorescence quantitative RT-PCR, the gene expression levels of N-myc, Fas, MTA1 and nm23-H1 in 30 cases of normal cervix organization, 30 cases of cervix invasive cancer (both for cervical squamous cell carcinoma) without metastasis and 30 cases with metastasis were detected and the results were analyzed. Results N-myc and MTA1 gene amplification levels in cervix invasive cancer tissue were higher than those in normal cervical tissue, the differences were statistical significant ( P 〈 0.05 ) ; Nm23-H1 and Fas gene amplification levels in cervix invasive cancer tissues werelower than those in normal cervical tissue, the differences were statistical significant (P 〈 0. 05) ; MTA1 gene amplification levels in cervix invasive cancer metastasis group were higher than those in the group without metastasis, nm23-H1 gene amplification levels were lower than that in not-metastasis group, the differences were statistical sig- nificant ( P 〈 0. 05 ) ; There were significant differences in N-myc, Fas gene amplification levels between cervix invasive cancer group and not-metastasis group ( P 〈 0. 05 ). Conclusions N-myc and MTA1 can promote cervical cancer cell proliferation, and Fas, nm23-H1 can restrain cancer cell proliferation. MTA1 gene can promote cervical cancer cell metastasis, and nm23-H1 gene can inhibit cervical cancer cell metastasis, and N-myc, Fas gene expression levels have correlation with cervical cancer cell proliferation and metastasis.
