电针对脑梗死大鼠神经功能及胶质纤维酸性蛋白表达的影响 被引量:7

Effect of Electroacupuncture on Neurological Function and Glial Fibrillary Acidic Protein Expression in Rats with Cerebral Infarction

摘要 目的观察电针对高血压基础上形成脑梗死大鼠的不同时间点神经功能评分及灶周胶质纤维酸性蛋白(GFAP)表达的影响,证实电针对星形胶质细胞(AST)变化的影响是其促进脑梗死功能恢复的机制之一。方法双肾双夹法制备RHRSP模型,双侧肾动脉狭窄术后12周,取血压≥l80mmHg大鼠,电凝法制备大脑中动脉闭塞(MCAO)模型,随机分为MCAO假手术组、MCAO组(模型组)、穴位治疗组、非穴位治疗组,后两组行电针刺激。MCAO后各组分别于1d、3d、7d、14d、21d进行神经功能缺损评分,HE染色观察病理形态变化,GFAP免疫荧光染色观察电针对局灶性脑梗死缺血灶周围GFAP的表达。结果假手术组大鼠在各时间点神经功能缺损评分均为0分;各时间点模型组神经功能评分均高于穴位治疗组,1d时两组差异无统计学意义,3d、7d、14d、21d,两组差异有统计学意义(P<0.05或P<0.01)。3d时模型组大鼠神经缺损症状最重,而电针组随着时间的延长其神经功能缺损评分在逐渐降低。在各时间点模型组与非穴位组无明显差异。HE染色可见穴位治疗组ASI及组织肿胀较模型组明显减轻。在五个时间点模型组和电针组GFAP表达都明显增高,与假手术组比较(P<0.01),电针组GFAP表达均明显低于同时段模型组(P<0.05)。各时间点模型组GFAP表达与非穴位组比较无显著性差异。结论穴位电针治疗能下调脑缺血后GFAP表达,进而改善神经功能。 Objective To observe the effect of electroacupuncture(EA) on neurological function and glial fibrillary acidic protein(GFAP) expression in rats with cerebral infarction.Methods Models were established by bilateral renal artery stenosis.After surgery for 12 weeks,rats with blood pressure ≥180 mmHg were used to prepare middle cerebral artery occlusion(MCAO) model,and randomly divided into MCAO sham operation group,MCAO group(model group),EA treatment group,and non-EA treatment group.After MCAO,neurologic impairment scores were observed.Pathological changes of GFAP were studied by immunofluorescence staining for focal CI around the ischemic lesions of GFAP expression(1 d,3 d,7 d,14 d,21 d).Results The neurologic impairment score was 0 in sham operation at each time point.At each time point,there was significant difference between the two groups in 3 d,7 d,14 d,21 d(P0.05 or P0.01).There was no significant difference in model group and the non-EA treatment group of all time points.Hematoxylin-eosin(HE) staining showed that astrocytes and tissue swelling were significantly reduced in EA treatment group compared with that in model group.GFAP expression in the five time points in model group and EA treatment group was significantly higher compared with that in sham group(P0.01).GFAP expression in EA group was significantly lower than that in model group at the same time(P0.05).Conclusion EA treatment could improve nerve function by lowering GFAP expression after cerebral ischemia.
