摘要
建立用高效液相色谱法测定血清中维生素A和维生素E的方法,并以之对某部新兵维生素A、E营养状况进行研究。血清样品加无水乙醇沉淀蛋白,上清液中加正己烷萃取维生素A、E,通氮气将正己烷提取液吹干后用甲醇溶解,进样分析。色谱柱为Shmadzu VP-ODS C18色谱柱(250mm×46mm,5μm),流动相为甲醇,维生素A和维生素E的检测波长分别为325nm和292nm。血清中维生素A、E分离良好,维生素A在90.6—1450.1μg/L范围内线性关系良好(r=0.9995),维生素E在4.7—75.1mg/L范围内线性关系良好(r=0.9998)。240名新兵血清中维生素A平均含量为321.217±54.356μg/L,维生素E平均含量为6.697±1.374mg/L。方法简便、快速、灵敏度高、准确性好,为进行营养状况监视和营养评价及干预提供了方便。
The method for the determination of vitamin A and vitamin E in serum by HPLC was established, and the situations of recruits' serum were researched. Vitamin A and vitamin E were extracted with hexane after the protein in serum was precipitated by absolute alcohol,and then the layer of hexane was dried by nitrogen. The residue that was dissolved by methanol was analyzed by HPLC. The HPLC system was consisted of Shmadzu VP-ODS C18 eolumn(250mm X 46mm, 5/;m) with the mobile phase of methanol,and the detection wavelengths of vitamin A and vitamin E were 325nm and 292nm,respectively. The results showed that vitamin A and vitamin E can separated,and linear ranges of vitamin A and vitamin E were 90. 6--1450. μg/L(r=0. 9995) and 4.7--75. 1mg/L (r=0. 9998),respectively. The average concentrations of vitamin A and vitamin E in recruits; serum were (321. 217±54. 356)μg/L and (6. 697±1. 374) mg/L. This method is simple,rapid with high sensitivity and accuracy, and provides convenience for nutriture monitoring, evaluation and intervention.
