摘要
[目的]优化发酵液中生化腐植酸(BHA)的提取工艺条件,并对其进行分析。[方法]运用单因素试验和正交试验优化BHA提取工艺;通过液相色谱、E4/E6、紫外-可见光谱、红外光谱对其进行定性、定量、芳香化程度、分子量、官能团种类及含量分析。[结果]最佳提取工艺条件为:提取液成分6 mol/L NaOH+0.025 mol/L Na4P2O7、提取液加入量17.5 ml、提取温度26℃、提取时间3 h,酸化pH为1.5,提取BHA量较优化前提高了29.17%;生化腐植酸和商品腐植酸结构相似,但BHA官能团种类较多、比例较高、分子量较小、芳香化程度较低。[结论]优化后的提取工艺合理可行,可用于提取生化腐植酸;试验所用4种分析方法简便快速,为生化腐植酸的结构研究、质量控制提供了依据。
[Objective] To optimum the extraction conditions of biotechnology humic acid and analyze it in different technologies.[Method] Extraction process of biotechnology humic acid was optimized by monfactorial experiments and orthogonal tests.Then biotechnology humic acid was characterized by HPLC,UV/VIS,E4/E6,IR.[Result] The optimal process was as follows: the biotechnology humic acid was extracted with 17.5 ml of the mixture of 6 mol/L NaOH and 0.025 mol/L Na4P2O7 solution for three hours in 26 ℃,then acidification to pH 1.5 with HCl.The results indicated that the biotechnology humic acid had more types of functional groups,a higher proportion,lower molecular weight and lower degree of aromatization than the commercial humic acid although they had the similar structure.[Conclusion] The extraction technology was reasonable and effective,which could be used to extract the biotechnology humic acid from fermentation liquor;four analytical methods were simple and rapid,which will provide a basis for structure research and quality control of the biotechnology humic acid.
