摘要
目的探讨苦参素对肝脏纤维化的抑制作用。方法利用四氯化碳制作大鼠肝纤维化和肝硬化动物模型,采用HE染色观察苦参素处理大鼠肝细胞结构的变化;通过Western blot实验观察肝组织中胶原Ⅰ,Ⅲ和基质金属蛋白酶MMP-13的变化。结果 HE染色结果 :CCl4组大鼠肝细胞溶解,弥漫性大片坏死,残留肝细胞成片气球样变,苦参素干预组大鼠的肝细胞结构形态明显好于CCl4组大鼠。Westernblot结果 :CCl4组大鼠的CollagenⅠ,Ⅲ的累积光密度与β-actin的比值比Con组分别提高了53.51%(P<0.01)和59.57%(P<0.01),MMP-13的累积光密度与β-actin比值降低了33.53%(P<0.01)。苦参素处理后CollagenⅠ,Ⅲ的累积光密度与β-actin的比值比CCl4组大鼠分别降低了26.66%(P<0.01)和30.75%(P<0.01),MMP-13的累积光密度与β-actin比值增加了38.43%(P<0.01)。结论苦参素可以抑制肝脏纤维化的进程。
Objective To investigate the inhibition effect of oxymatrine on hepatic fibrosis. Methods The rat models with hepatic fibrosis and liver cirrhosis were established by carbon tetrachloride ( CC14 ), the changes of structure of hepatocyte were observed by HE staining, and changes of expression of collagen I , Ⅲand MMP-13 were observed by means of Western Blot. Results HE staining results:the hepatolysis, diffuse necrosis and ballooning degeneration of remained hepatocytes were found in CC14 group, however, the structure and morphous in oxymatrine intervention group were superior to those in CC14 group. Western Blot results:the ratio of accumulation OD of collagen β-actin and collagen Ⅲ/β-actin in CC14 group was increased by 53.51% ( P 〈 0.01 ) and 59.57% ( P 〈 0.01 ), as compared with that in control group, however, the ratio of accumulation OD of MMP-13/β-actin was decreased by 33.53% ( P 〈 0.01 ). After treated by oxymatrine, the ratio of accumulation OD of collagen I/β-actin and collagen Ⅲ/β-actin in CC14 group was decreased by 26.66% ( P 〈 0.01 ) and 30.75% ( P 〈0.01 ) ,as compared with that in control group,however,the ratio of accumulation OD of MMP-13/ β-aetin was increased by 38.43% ( P 〈 0.01 ). Conclusion Oxymatrine can inhibit the process of hepatic fibrosis.
