摘要
目的研究不同地理来源的食品中大肠埃希氏菌的群体进化关系,为流行病学调查及有效控制食源性大肠埃希氏菌导致的食品安全事件提供理论支持。方法按照大肠埃希氏菌多位点序列分型(multilocus sequence typing,MLST)规则,根据地理溯源研究中遴选的4个持家基因测序数据,利用数据库查询每株食品分离株的序列类型(sequence types,ST),再对所有食品分离株进行eBURST(enhanced based upon related sequence types)分析。结果得到了食品中大肠埃希氏菌分离株群体的遗传关系。eBURST分析显示不同地理来源菌株存在相同起源株,不仅在同一大洲相邻国家或地区之间,而且在不同大洲非相邻国家或地区之间都存在着同质性的克隆株复合体。结论大肠埃希氏菌在具有相同起源的基础上存在丰富的遗传多样性;eBURST分析更擅于揭示不同地理来源大肠埃希氏菌株之间的进化关系。
Objective To analyze the population evolutionary relationship among Escherichia coli strains from food with different gepgraphic sources by enhanced based upon relatad sequence types(eBURST) analysis,so as to provide theoretical basis for epidemiologic survey and effective control of food safety event induced by E.coli.Methods A total of 253 Escherichia coli strains obtained from food were analyzed by eBURST based on sequences of housekeeping genes from the E.coli multilocus sequence typing(MLST) database.Results eBURST analysis showed that E coli strains from different geographic sources are of the same origin strain.Homogeneous clonal complex not only exists in the neighbouring countries or regions,but also in non-neighbouring countries in different continents.Conclusion E coli strains have great genetic diversity in spite of the same origin.eBURST analysis is good at revealing the evolutionary relationship among E coli strains from different geographic sources.
