摘要
目的建立高效液相色谱法测定妇炎膜中苦参碱及氧化苦参碱的含量。方法采用Hypersil NH2(4.6 mm×250 mm,5μm)色谱柱,以乙腈-无水乙醇-3%磷酸溶液(85:7.5:7.5)为流动相,柱温30℃,检测波长为220 nm,流速1.0 mL/min,理论塔板数按氧化苦参碱计算,不低于5 000。结果苦参碱在0.632 4μg^3.4620μg范围内线性关系良好,r=0.999 7,平均加样回收率为101.29%,RSD=1.41%:氧化苦参碱在0.070 8μg^0.354 0μg范围内线性关系良好,r=0.999 9,平均加样回收率为96.38%,RSD=0.87%。结论该方法简便准确,灵敏可靠,专属性好,重现性好,可以作为妇炎膜中苦参碱及氧化苦参碱的含量测定的方法。
Objective To establish a HPLC method for determination of matrine and oxymatrine in Fuyan membrane.Methods Using Hypersil NH2(4.6 mm×250 mm,5μm)column,acetonitrile-ethanol-3% phosphoric acid(85:7.5:7.5) as mobile phase,column temperature was 30 ℃,detection wavelength was set at 220 nm,flow rate was 1.0 mL/min,theoretical plate numbers calculated by oxymatrine was not less than 5000.Results The linear range of matrine was 0.632 4 μg ~ 3.462 0 μg(r= 0.999 7),the average recovery was 101.29% with RSD =1.41%.The linear range of oxymatrine was 0.070 8 μg ~ 0.354 0 μg(r= 0.999 9),the average recovery is 96.38% with RSD = 0.87%.Conclusion This method was simple,accurate,sensitive and reliable,and could be the determination method of matrine and oxymatrine in Fuyan membrane.
