摘要
树突状细胞(DC)是高效的抗原呈递细胞,有启动初始免疫应答的独特功能。未成熟或亚成熟DC来源于CD34+的造血祖细胞,通过吞噬、吞饮的方式摄取抗原,进而成熟转变为更有利于呈递抗原的成熟DC。成熟的DC将抗原呈递给CD4+的Th细胞,激活T细胞、B细胞、巨噬细胞和自然杀伤细胞,进行特异性的免疫应答。DC呈递功能高效性的疫苗疗法逐渐加入到抗肿瘤及抗各种感染性疾病治疗的行列中。从实验室到临床应用,DC的疫苗制备及抗原的负载方法不断衍生出更高效、严格的技术方法。
Dendritic cells(DC)are highly-efficient antigen-presenting cells with a unique ability to induce primary immune responses.DC progenitors with high phagocytic capacity were expanded from CD34 + hemopoietic progenitor cells.Immature DCs capture Ag and subsequently migrate to the lymphoid organs and present Ag to CD4 + T-helper cells,which in turn regulate the immune effectors,including T cells and B cells,as well as macrophages and NK cells.New ways of anti-neoplastic and anti-infective were found based on the efficient antigen-presentation of DCs.More efficient and strict technical methods of DC vaccine preparation and antigen load are continuously derived,from the laboratory to the clinical application.
