摘要
目的探讨缝隙连接(GJ)在阿尔茨海默病(AD)发病中的作用,研究尼莫地平对于认知功能的影响及对脑内缝隙连接蛋白(Cx)的调节作用。方法采用脑室内注射Aβ25-35制备的大鼠阿尔茨海默病模型,利用Y迷宫检测大鼠的学习记忆功能,研究尼莫地平对于认知功能的影响,采用免疫印迹方法检测大鼠海马Cx43的表达水平。结果同AD组相比,尼莫地平治疗大鼠Y迷宫中正确反应次数增多,说明尼莫地平可改善AD大鼠的学习记忆功能。与对照组相比,AD大鼠海马Cx43蛋白表达增高,而尼莫地平可以降低AD大鼠海马Cx43表达水平。结论钙通道阻滞剂尼莫地平可以改善AD大鼠的学习记忆功能,这可能与其影响海马Cx43表达水平有关。
Objective To study the effect of nimodipine on learning and memory of rat model of Alzheimer's disease(AD) and to explore its effect on the expression of connexin 43(Cx 43).Methods 30 Sprague-Dawley rats were selected and randomly divided into 3 groups: control,AD,and nimodipin group.The rat AD model was established by stereotactically injecting amyloid beta protein(Aβ) 25-35 into the bilateral ventricles.Learning and memory functions were tested using the Y-type electric maze.The expression of hippocampal Cx43 was tested by western blot.Results Compared with the AD group,the number of correct reaction in Y maze increased after nimodipine treatment.Nimodipine could decreased the Cx 43 expression which was proved to be elevated in hippocampus of AD rat.Conclusion Nimodipine can improve the learning and memory ability of AD rat,which may be related to its downregulation of Cx43.
