摘要
目的:观察苦参碱对豚鼠心室肌动作电位及钠通道的作用,探讨其抗心律失常的作用机制。方法:采用标准微电极记录技术记录动作电位,采用膜片钳全细胞记录技术记录通道电流。结果:苦参碱(10μmol/L;50μmol/L;100μmol/L)浓度依赖性延长APD50和APD90,降低APA(n=8);乌头碱明显延长APD50和APD90,苦参碱(100μmol/L)能逆转乌头碱对动作电位的过度延长作用(n=7);苦参碱(50μmol/L)抑制IN a,与对照组相比,其IN a电流密度在-45mV时从(-65.14±4.2)pA/pF降低到(-46.39±4.8)pA/pF(n=10)。结论:苦参碱能逆转乌头碱对动作电位时程的延长作用并能抑制IN a。
Objective:To observe the effect of Matrine on action potential and sodium channel in guinea pig papillary muscles thus to clarify its mechanisms of antiarrythmic effects.Methods:Conventional microelectrode technique was applied to record the action potential and the whole-cell patch clamp recording technique was used to record the change of sodium channel current.Results Matrine prolonged the action potential duration at 50%,90% repolazation(APD50,APD90) in a dose-dependent manner(P0.01,n=8) and decreased APA(P0.05,n=8);Aconitine significantly prolonged the action potential.Matrine(100μmol/L) reversed action potential duration induced by the aconitine(n=7).Matrine(50μmol/L) decreased the sodium channelcurrent density from(-65.14±4.2)pA/pF to(-46.39±4.8) pA/pF(-45mV)(n=10).Conclusion Matrine reversed the changes of action potential duration induced by aconitine and decreased sodium channel current.
