山楂叶总黄酮对大鼠心肌缺血性损伤的保护作用及机制研究 被引量:25

Protective effect and the mechanisms of Harthorn leaves flavonoids on myocardial ischemia in rats

摘要 目的:探讨山楂叶总黄酮对大鼠心肌缺血性损伤的保护作用及通过HO-1诱导途径的作用机制。方法:建立皮下注射异丙肾上腺素造成大鼠心肌缺血模型,观察山楂叶总黄酮对心电图、血清肌酸激酶CK)和乳酸脱氢酶(LDH)活性、心肌超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)的活性以及血红素氧合酶-1(HO-1)蛋白表达和mRNA水平的影响。结果:山楂叶总黄酮(100~200mg/kg)能显著对抗异丙肾上腺素所致急性大鼠心肌缺血心电图T波的异常,显著降低血清LDH和CK活力,增加心肌组织SOD和GSH活性,上调HO-1的表达,并呈一定的剂量依赖性。HO-1抑制剂组能够部分取消HLF的对异丙肾上腺素造成的心肌损伤的保护作用。结论:山楂叶总黄酮对急性缺血性心肌损伤的保护作用机制与诱导HO-1的表达密切有关。 Objective: To investigate the protective effect of hawthorn leaf flavonoids on isoprenaline induced acute myocardial iscbcmia and the mechanisms with HO-I in rats. Methods :The healthy male SD rats were treated with HLF (100mg/kg and 200mg/kg, ip) for 5 days. The acute myocardial ischemia models were induced by subcutaneous injection of isoprenaline (Smg/kg, once a day, and lasted for 2 days) . HO-1 inhibitor (Znpp) were interrupted the effect of HO-1. Results :Compared with the model group, HLF could effectively ameliorate Twave change induced by isoprenaline, significantly decrease serum CK an LDH enzymic activity, and distinctly increase the level of SOD and GSH in myocardial tissue, and significantly enhance the expression of HO-1 and upregulate its mRNA. However, those effects can be inhibited by Znpp. Therefore, the protective effect of HLF on acute myocardial ischemia induced by isoprenaline in rat may related to HO-1.
