摘要
目的观察异丙嗪和恩氟烷合用对小鼠的镇痛和催眠效应。方法72只小鼠随机分为3批,每批24只,分别用于热板实验、扭体实验和催眠实验。每批小鼠再随机分为4组(每组n=6):生理盐水组(NS组)、异丙嗪组(P组)、恩氟烷组(E组)及异丙嗪恩氟烷合用组(PE组)。通过热板实验和扭体实验分别观察各组小鼠的热板痛阈(HPPT)、扭体潜伏期和扭体次数(WT);通过催眠实验观察各组的睡眠潜伏期和睡眠时间(ST)。结果热板实验结果表明:各组小鼠基础HPPT差异无显著性,异丙嗪和恩氟烷单独给药后HPPT与基础HPPT相比无显著变化,而二者合用后各时间点HP门均升高(P〈0.01);在同一时间点与NS组比较,异丙嗪和思氟烷单独给药不能显著改变小鼠的HPPT;而二者合用后,与NS组、P组、E组比较HPPT均显著升高(P〈0.05或P〈0.01)。扭体实验结果表明:异丙嗪单独应用既不能延长扭体潜伏期,也不能减少小鼠的扭体次数;恩氟烷单独应用仅能够延长扭体潜伏期(P〈0.01)而不能减少扭体次数;二者合用后,与NS组、P组、E组比较小鼠的扭体潜伏期延长和扭体次数减少(P〈0.01)。催眠实验结果显示:异丙嗪和恩氟烷单独使用无小鼠入睡,而二者合用后小鼠全部入睡。结论异丙嗪和恩氟烷联合用药可产生协同的镇痛和催眠效应。
Objective To observe the analgesic and hypnotic effects of promethazine combined with enflurane in mice. Methods A total of 72 mice were randomly divided into three batches. There are 24 mice in each batch, respec- tively for hot - plate, writhing and hypnotic tests. Each hatch of mice were then randomly divided into four groups ( n = 6 each) : normal saline group (NS) , promethazine group (P) , enflurane group (E) , combination of promethazine and en- flurane group (PE). HPPT, writhing times and writhing latency of mice were respectively observed in each group through hot - plate test and writhing test. Sleeping time and sleeping latency were also respectively observed in each group through hypnotic test. Results Hot - plate test showed that there was no significant difference in basal HPPT of mice among groups. After separate injection of pomethazine or enflurane, the HPPT of mice was not significantly changed compared to basal HPPT. But HPPT of mice was significantly enhanced at each time point after pomethazine and enflurane combined administration ( P 〈 0.01 ). Compared with NS group at the same time points, HPPT of mice was not changed significant- ly through injecting mice with pomethazine or enflurane. But after combined administration of them, HPPT of mice was significantly enhanced compared with NS, P and E group (P 〈 0.05 or P 〈 0.01 ). In writhing test, it was showed that pomethazine neither lengthen writhing latency nor decrease writhing times of mice. Enflurane could lengthen writhing la- tency (P 〈 0.01 ) and hardly decrease writhing times of mice. Pomethazine and enflurane combined administration could significantly lengthen writhing latency and decrease writhing times ( P 〈 0.01 ). In hypnotic test, it was showed that sepa- rate injection of pomethazine or enflurane could not shorten sleeping latency and lengthen sleeping time, while combina- tion of pomethazine and enflurane could significantly shorten sleeping latency and prolong sleeping time compared withNS, P and E group (P 〈 0. 01 ). Conclusion Combined injection of pomethazine and enflurane can produce synergistic effects of analgesia and hypnosis.
