摘要
目的利用电子自旋共振(electron spin resonance,ESR)波谱仪检测肝纤维化过程中血清、血浆以及肝脏中分别产生的自由基信号,寻找检测自由基的样品前处理最佳方案。方法采用传统的四氯化碳法造模,造模1周后通过不同的处理方法,利用ESR检测小鼠肝脏、血清和血浆的自由基信号。结果通过利用ESR技术,在小鼠肝脏、血清及血浆中均检测到了自由基信号,并且在处死小鼠后加入电子捕获剂(DMPO)所测得的自由基信号更加明显。结论通过以上对比发现,样品加DMPO的生理盐水组测出的自由基信号相较于其他处理方法而言更为明显。
Objective To detect free radical signals of blood serum,plasma as well as the liver in the procession of fibrosis by using ESR,in order to find the best approach of dealing with sample before detecting.Methods Taking advantage of traditional method by which model was made by CCl4,free radical signals of blood serum,plasma as well as the liver were detected by ESR technology one week later via different sample ways.Results Utilizing ESR,free radical signals has been detected in rats' liver serum and plasma.In addition,the groups which added DMPO could detect more obvious free radical signals compared with other groups.Conclusion According to this method,the samples which was added DMPO and then treated with saline water coulde be detected obvious signal in contrast to other ways.
