中国姥鲨软骨源新生血管抑制因子(Sp8)的分离纯化及生物学活性 被引量:15

摘要 目的 :从中国姥鲨软骨组织抽提蛋白质中筛选具有新生血管抑制活性和抗肿瘤活性的分子。方法 :盐酸胍抽提姥鲨软骨蛋白 ,采用超滤和分子筛柱层析等方法分离纯化获得相对分子质量为 8.3× 10 3的蛋白质 (Sp8) ,以体外抑制血管内皮细胞增殖、体内抑制新生血管生长和抑制小鼠移植 S180肉瘤生长作为筛选指标。 结果 :Sp8可在体外抑制血管内皮细胞增殖 ,40μg/ml浓度时抑制率为 2 0 % ;在体内具有抑制新生血管生长的作用 ,兔角膜模型抑制率为 72 .7% ,鸡胚模型 (CAM)抑制率为 79% ;并能明显抑制小鼠体内移植 S180肉瘤的生长 (P<0 .0 0 1)。结论 :Sp8介导了鲨鱼软骨中的抗肿瘤活性 ,其作用可能是通过抑制肿瘤诱生新生血管形成这一途径来实现的。 Objective: To screen shark cartilage derived active proteins which can inhibit angiogenesis and tumor growth. Methods: Shark cartilage derived proteins were extracted by 1 mol/L guanidine hydrochloride. Highly purified Sp8 was obtained by ultrafiltration and Superdex 75 column chromatography. The activities of Sp8 were studied on its inhibition of vascular endothelial cell growth, angiogenesis both on rabbit cornea and chick embryo chorioallantoic membrane (CAM) assay, and growth of transplanted S180 sarcoma in mice. Results: (1) Sp8 could inhibit the growth of vascular endothelial cells in vitro . At the concentration of 40 μg/ml, the inhibitory rate was 20%. (2) Sp8 showed strong inhibition of angiogenesis, the inhibitory rate reached 72.7% on rabbit cornea and 79% on CAM model, respectively. (3) Sp8 could efficiently reduce the mass of S180 sarcoma growing in mice, IC50 on this model was less than l00 μg/kg. Conclusion: Sp8 mediates at least part of the anti tumor activity endowed by shark cartilage and may work through the inhibition of tumor induced angiogenesis.
