摘要
目的建立测定姜黄素血药浓度的方法。方法采用高效液相色谱-质谱联用法对姜黄素的血药浓度进行测定,采用固定相为Hypersil GOLD C18柱,以甲醇-水为流动相,进行梯度洗脱,流速为0.2mL.min-1,柱温为35℃;质谱采用高温电喷雾离子源,以选择反应监测模式进行定量分析,血浆样品用乙腈直接沉淀处理。结果姜黄素在2.5~187.5ng.mL-1范围内呈良好的线性关系。日内、日间RSD均小于15%(n=5)。结论本方法操作简单、灵敏度高、分析时间短,适用于姜黄素血药浓度监测及药代动力学研究。
Objective To establish an LC-MS/MS method to determine the content of curcumin in plasma.Methods A high performance liquid chromatography-electrospray mass spectrometry method was utilized for rapid determination of curcumin in plasma.The analysis was carried on a Hypersil GOLD C18 column.The column temperature was 35 ℃,with linear gradient elution using water and methanol,and the flow rate was 0.2 mL·min-1.The quantitative analysis was done by selected reaction recording(SRM)mode of heater electrospray ionization(HESI).The plasma sample was deposited by acetonitrile directly.Results A good linearity was detected in the linear range of 2.5-187.5 ng·mL-1 for curcumin;both intra-day and inter-day RSD were less than 15%(n=5).Conclusion The method is simple,sensitive,rapid and suitable for therapeutic drug monitoring and pharmacokinetic study.
